Delivery of chromatographic workstation

Lot 1: Chromatographic workstation

The chromatographic workstation should be composed of an analytical and a preparative HPLC, which are controlled by a common PC incl. software and which can share a mass selective detector (MSD) (in the respective device configuration). The following workflows should be realized:

1.Separation of complex extracts from plants, algae, fungi and bacteria on an analytical scale and detection of the separated compounds by means of diode array detector (DAD) and/or mass selective detector (MSD) as an overview run.

2.Software-assisted optimization of the separation of analytical HPLC runs for the extraction of individual substances based on the signal of the DAD and/or MSD. Subsequent software-based upscaling to the preparative scale with method for fractionation by wavelength by means of multi-wavelength detector and/or by mass by MSD.

3.Recovery of the separated components after upscaling on a preparative scale with the possibility of collection of individual components by wavelength by multiwavelength detector and/or by mass by MSD.

4.Possibility of fractionation into different vials adapted to the expected fraction size and subsequent injection from the same vials to check the purity of the fractions obtained, thus requiring a device that works both as a fraction collector and as a sampler.

5.Use of analytical HPLC as a high-end device with DAD and MSD. This requires an analytical thermostatizable autosampler with high accuracy and precision injection volume for low volumes as well as the possibility of temperature control of the analytical columns by means of a column thermostat.

6.Rapid optimization of the separations in terms of selectivity due to the possibility of column selection by column switching valve on both analytical and preparative scale.

7.Control of HPLC by chromatography software, whereby a separate configuration is required for the analytical mode and the preparative mode of operation. By means of signals programmed by the user (triggers), the fractionation of the chromatographic runs as well as the collection of corresponding fractions should be carried out automatically. The PC required for this must also be equipped with software for evaluating the chromatograms.

As accessories for the safe operation of the chromatographic workstation, 3 safety cabinets are required:

1.Working on a preparative scale requires the provision of large amounts of solvent as eluents (at least 20 L per eluent at a binary gradient for an 8-hour working day) due to the higher flow rates. For this purpose, storage is provided in a safety cabinet, which must have professionally mounted bushings for the connecting cables to the pump that meet the safety requirements and whose installation is therefore intended directly next to the chromatographic workstation. (Table 1)

2.When the chromatogram is "cut" into fractions, larger amounts of waste are produced at the same time in addition to the desired fractions. This is all the higher if only one or two substances are specifically fractionated. Therefore, a second safety cabinet is required, which also collects the waste in 20 l canisters. (Table 2)

3.The collected fractions, which often contain light- and heat-sensitive substances, must be stored in a cool and dark place. Therefore, a cooled safety cabinet is required as the 3rd safety cabinet for storing the fractions. (Table 3)

4.Canisters with safety waste caps are necessary for the submission or absorption of larger quantities of solvent.
