This public tender process is for oligos and probes for HUS Diagnostic Center, Laboratory of Genetics at Helsinki University Hospital.

The acquired oligos and probes are used to study genetic alterations, e.g. point mutations, tandem repeats, fusion transcripts, copy number changes in both somatic and congenital diseases. The combination of fluorochromes allows different alterations to be explored simultaneously. In the laboratory, the most common methods that require oligos and probes are Sanger sequencing, cyclic minisequencing, quantitative PCR and fragment analysis. Approximately 50,000 genetic tests are performed each year.

The tender is devided into three lots:

1) Standard oligos

2) Probes and oligos

3) Customized oligos and probes

The procurement entity selects one supplier for each lot so that the lots may have either the same or a different supplier.

This lot applies to standard oligos, most of these are in use or new, patient or family specific oligos. Standard oligos are ordered weekly.

Standard oligos should also be available with different modifications at the 5’ or 3' ends or both.

The fluorochromes in the modifications must match the settings for the excitation and emission filters of equipment used in the laboratory.

Delivery time for oligos without modification is a maximum of 5 working days from the order. Delivery time for modified oligos is up to 10 working days from the order.

This lot applies to probes that contain both oligos and a probe. In the laboratory, dual-coloured probes based mainly on TaqMan chemistry are used for quantitative PCR.

Probes should be available with different modifications at the 5 'and 3' ends.

The fluorochromes in the modifications must match the settings for the excitation and emission filters of equipment used in the laboratory.

The delivery time of the probes is a maximum of 15 working days from the order.

The requirements are further specified in the call for tenders and its annexes.

This lot applies to customized oligos and probes designed and pre-validated by the manufacturer.

The manufacturer is given either a gene with a mutation according to the genetic nomenclature, e.g. BRAF c.1799T> A p. (Val600Glu) or by presenting the sequence of the region. The functionality of the designed oligos and probes must be verified.

Oligos and probes should be available with different modifications at the 5 'or 3' ends or both.

The fluorochromes in the modifications must match the settings for the excitation and emission filters of equipment used in the laboratory.

Delivery time for designed and pre-validated oligos and probes is up to 30 working days from order.
