EXPLORATORY NOTICE IN ORDER TO ACQUIRE EXPRESSIONS OF INTEREST FOR THE SUPPLY IN SERVICE OF A "NEXT GENERATION SEQUENCING, NGS" SEQUENCING SYSTEM NEEDED BY ASST FATEBENEFRATELLI SACCO
supply in service of a "Next Generation Sequencing, NGS" sequencing system, for an indicative duration of 36 months, possibly renewable for a further 36 months, including:
• Instrumentation for massively parallel sequencing "Next Generation Sequencing, NGS" automated, high throughputivity, with sequencing chemistry by synthesis and wide range of sequencing applications.
• Technical assistance and ordinary and extraordinary maintenance service, guaranteed for the entire duration of the supply both with on-site interventions and with telephone contact.

• Reagents for the preparation of libraries for NGS from nucleic acids of different pathogens (viruses, bacteria, fungi, parasites) extracted from biological samples of different matrices
• Sequencing reagents and consumables
• Fluorimeter with dedicated reagents and consumables
• Reagents for purification with magnetic marbles
• Mini centrifuge for test tubes (up to 2.0 mL, maximum speed required 12,000 x g)
• Mini-centrifuge for 96-well plates
• Magnetic holder for test tubes for the purification of amplicons (1.5 mL)
• Set of single-channel and multi-channel micropipettes with adjustable volume
• High mixer with adjustable speed (up to 3,000 rpm)
• Automatic software for the analysis and interpretation of specific NGS data
The NGS sequencer must have the following minimum characteristics:
• output da 500 Mb a 8 Gb
• sequencing run readings: 2 to 30 million
• Reading length: 2 × 150 bp to 2 × 250 bp
The consumables required for the preparation of genomic libraries must have the following minimum characteristics:
1) Unique CE-IVD kit for the evaluation of drug resistance and tropism of HIV-1 (sequencing of protease regions, retrotranscriptase, integrase and V3 loop of HIV-1) (300 reactions/year)
2) Kit for the evaluation of HCV drug resistance and the determination of the viral genotype (50 reactions/year)
3) Kit for the evaluation of HBV drug resistance and the determination of the viral genotype (50 reactions/year)
4) kit for full-genome sequencing of SARS-CoV-2 for the determination of viral variants (200 reactions/year)
5) kit for sequencing a panel of respiratory viruses (100 reactions/year)
6) open-label sequencing kit with metagenomic approach (50 reactions/year)
7) Kit for the preparation of NGS libraries of pathogens (bacteria and fungi), starting from ≤10 ng of DNA extracted from biological samples of different matrices, including fresh tissues, cytological samples, plasma. Also, optional matrix: FFPE. (200 reactions/year)
